Summary

Retatrutide is a triple agonist at GLP-1, GIP, and glucagon receptors designed to combine appetite suppression with increased energy expenditure and hepatic fat oxidation. Phase 2 data show very high average weight loss compared with GLP-1 or dual agonists, but with a higher rate of some adverse events. It remains investigational, and long-term safety and cardiovascular outcomes are still being defined in ongoing trials.
